ABSTRACT

ABSTRACT Foliar diseases impose severe restrictions on the persistence and productivity of Medicago sativa, both of which may be increased by developing disease resistant and more competitive genotypes that can improve pasture quality. We found Curvularia geniculata as the principal alfalfa foliar pathogen in the Brazilian state of Rio Grande do Sul (RS). Growth chamber experiments evaluated the resistance of alfalfa genotypes 'E1C4', 'CPPSul', 'ABT 805' and 'CUF-101' to C. geniculata as compared the control 'Crioula' genotype. These genotypes were also evaluated in field trials at a sea level site in Eldorado do Sul in central RS and at two sites £200 m above sea level, one in Bagé municipality in south west RS and the other at a farm near the town of Alto Feliz in north east RS. Plants were spray-inoculated with 1.6 x 106 ml-1 of C. geniculata spores and visually evaluated for leaf damage 14 days later. The C. geniculata infection rates varied from zero to 100%. Alfalfa persistence and forage mean dry mass (DM) production at the Eldorado site were measured during different seasons from November 2013 to January 2015 by calculating the incidence of invasive plants and morphologically separating leaves from stems and calculating both leaf and stem DM. Data were analysed using mixed statistical models. The best results for persistence and forage DM were shown by the 'CPPSul' genotypes (DM = 16,600 kg ha-1) and 'Crioula' (DM = 15,750 kg ha-1). These two genotypes will be used for subsequent investigations and selection cycles.

ABSTRACT The Paspalum genus includes several species that are important for livestock in Rio Grande do Sul, such as P. notatum and P. guenoarum, typical of native pastures of the Pampa biome. The aim of this study was to investigate forage production and chemical composition of four ecotypes of these species in relation to the cv. 'Pensacola' (P. notatum). Ecotypes of P. guenoarum (Azulão and Baio) and P. notatum (André da Rocha and Bagual) and the cv. 'Pensacola' were evaluated for two years, during which four cuts/year were made. The work was carried out under field conditions at the Agronomic Experimental Station of the Federal University of Rio Grande do Sul (30°05'S; 51°39'W), in a completely randomized design. P. guenoarum stood out for higher productivity and greater tolerance to cold; the Azulão ecotype showed more autumn production in relation to the other ecotypes. Crude protein content ranged from 14 (Baio) to 15% ('Pensacola'); for neutral detergent fiber, the variation was 68 (Azulão) to 71% ('Pensacola') and for acid detergent fiber there was a variation of 38 ('Pensacola') to 43% (Baio). The data demonstrates the potential of native genotypes for use as cattle feeding in southern Brazil.

Abstract Aiming to investigate new therapeutic agents with fewer side effects, the number of studies about natural products has increased. Phenolic compounds comprise a well-studied class of abundant plant-derived compounds, whose anti-inflammatory activity has been described. Isoflavones are phenolic compounds that occur mainly in the Leguminosae family, and can be found in many species, such as Trifolium riograndense Burkart, Leguminosae (clover). In this study an HPLC method was used to determine and quantify four isoflavones (genistein, daidzein, formononetin, and biochanin A) in hydrolyzed leaf, flower, stolon, and root extracts of T. riograndense. In vivo anti-inflammatory activity was investigated using the rat paw edema method and in vitro chemotaxis model with a dry extract from the leaves, which had the highest amount of isoflavones. The major isoflavone found in all parts of the plant was formononetin. The chemotaxis assay revealed that the different concentrations (0.2-50 µg/ml) of the dry extract significantly inhibited neutrophil migration in a concentration-dependent manner (more than 90%). In the rat paw edema test, oral administration of clover extract 100 mg/kg was able to significantly inhibit the edema formation induced by carrageenan. In conclusion, chemical analyses showed that Trifolium riograndense is a plant rich in isoflavones and a new interesting option as isoflavone source. The results of the biological tests taken together show that the extract of T. riograndense has anti-inflammatory effect in rodents.

ABSTRACT: The goal of this study was to assess the variability of Paspalum plicatulum x P. guenoarum interspecific hybrids in terms of agronomic performance and their tolerance to cold conditions, as well as to estimate the correlation of different phenotypic characters associated with forage production. Twenty hybrids plants were used, besides one access P. guenoarum , one of P. plicatulum and the cultivar 'Pensacola' ( P. notatum ). Assessments were performed on individual plants, in a completely randomized design with five repetitions. Paspalum genotypes presented variability in total dry matter, leaf dry mass, leaf: stem ratio, and tolerance to cold. We suggest selection of the hybrids 08Q01 and 08Q44 for new stages within a forage improvement program. Estimates of phenotypic correlation may aid in the selection of genotypes with better agronomic traits.


RESUMO: O objetivo deste estudo foi avaliar a variabilidade de híbridos interespecíficos de Paspalum plicatulum x P. guenoarum por meio do desempenho agronômico e tolerância ao frio, bem como estimar as correlações fenotípicas de diferentes caracteres ligados à produção de forragem. Foram utilizados 20 híbridos, um acesso de P. guenoarum , um de P. plicatulum e a cultivar 'Pensacola' ( P. notatum ). As avaliações foram realizadas em plantas individuais, em delineamento completamente casualizado, com cinco repetições. Os genótipos de Paspalum apresentam variabilidade para massa seca total, massa seca de folhas e relação folha: colmo e na tolerância ao frio. É possível selecionar os híbridos 08Q01 e 08Q44 para novas etapas dentro de um programa de melhoramento de forrageiras. As estimativas de correlações fenotípicas podem auxiliar na seleção de genótipos com caracteres agronômicos superiores.

ABSTRACT

The objective of this study was to analyze the adaptability and the stability in apomictic genotypes of the genus Paspalum across the main characters of forage interest evaluated in different years and locations. The experiment was conducted in the years 2010 and 2011 in the municipalities of Eldorado do Sul and Augusto Pestana, RS, Brazil, in a randomized complete block design with three replications. Analyses of adaptability and stability were calculated by the Traditional and the Eberhart and Russel method. The genotypes of P. guenoarum Baio anda Azulão shows high biomass production but adapted to favorable environments and reduced stability. The genotypes of P. lepton 28C show the total dry mass specific adaptability to unfavorable environment and in the dry leaf adaptability with stability.


O objetivo do trabalho foi analisar a adaptabilidade e estabilidade de genótipos apomíticos do gênero Paspalum frente aos principais caracteres de interesse forrageiro avaliados em diferentes anos e locais. O experimento foi conduzido nos anos de 2010 e 2011 nos municípios de Eldorado do Sul e Augusto Pestana, RS, Brasil, em um delineamento experimental de blocos casualizados com três repetições. As análises de adaptabilidade e estabilidade foram calculadas pelo método Tradicional e pelo Eberhart e Russel. Os genótipos de P. guenoarum Baio e Azulão mostram alta produção de biomassa, mas ajustados a ambientes favoráveis e com reduzida estabilidade. O genótipo de P. lepton 28C evidencia, na massa seca, total adaptabilidade específica a ambiente desfavorável e, na massa seca de folha, adaptabilidade geral com estabilidade.

ABSTRACT

Red clover is one of the most utilized forage in agriculture and contains many of the isoflavones known for their human health benefits. The objectives of this study were: i) to quantify, using HPLC analysis, isoflavones in 77 accessions from the USDA core collection and a Brazilian line; ii) to verify possible relationships depending on their origin, improvement status or maturity type and; iii) to verify the seasonal variation. The isoflavone mean contents were 29.27 µg g-1 of dry material for daidzein, 163.69 µg g-1 for genistein, 11353.29 µg g-1 for formononetin and 6568.8 µg g-1 for biochanin A. Clustering was mainly influenced by the total amount of isoflavones and partially due to maturity type, improvement status and geographic origin. The seasonal evaluation demonstrated an increase of concentration during winter, and decrease during spring. These results highlighted accessions that can be used to develop new varieties with low or high isoflavones concentration.

ABSTRACT

Red clover Trifolium pratense L., Fabaceae, contains four isoflavones, mainly formononetin and biochanin A, and in smaller concentrations, daidzein and genistein. These compounds have gained a lot interest due to its human health benefits, such as estrogenic and progestogenic activities, antioxidant, anti-cancer and others. The objective of this study was to determine in vivo and in vitro anti-inflammatory activity of red clover dry extract. The in vitro anti-inflammatory activity was assayed by the technique using the Boyden chamber method, evaluating the leukocyte migration inhibition (chemotaxis). The in vivo anti-inflammatory activity was tested by a carrageenan-induced rat paw edema test. The results of anti-inflammatory in vitro test showed that there was a significant inhibition of leukocyte migration at the concentrations of 100, 50, 25, 10 and 5 µg/mL of red clover dry extract, these doses resulted in 94.73, 95.39, 94.73, 84.68 and 78.75 percent of inhibition for each dose, respectively. The anti-inflammatory in vivo test resulted in a significant activity in both tested doses (100 and 50 mg/kg of red clover dry extract) and at each tested time. The average percentage of edema inhibition was 63.37 percent. The findings of this study suggested that red clover extract might be suitable for the treatment of inflammatory diseases.

ABSTRACT

O trevo branco é uma leguminosa forrageira muito importante para utilização em pastagens temperadas no Rio Grande do Sul. Entretanto, ele é mais sensível ao déficit de água no solo do que as outras leguminosas perenes, apresentando problemas de persistência no verão. Com o objetivo de determinar características morfofisiológicas relacionadas à resposta ao déficit hídrico dessa espécie, foi realizado um experimento em casa-de-vegetação, o qual avaliou o efeito da disponibilidade hídrica (90 e 40 por cento da umidade de capacidade de campo do solo) sobre sete acessos pertencentes à Coleção Nuclear de Trevo Branco do Departamento de Agricultura dos Estados Unidos (USDA). O delineamento experimental utilizado foi inteiramente casualizado, com quatro repetições, totalizando 56 vasos. Foram realizadas duas avaliações, aos 130 e aos 196 dias após a semeadura, ocasião em que foram avaliadas diversas características morfofisiológicas. A análise dos resultados indicou que o déficit hídrico teve um efeito marcante sobre a altura de plantas (EST), área foliar (AF), número de folhas vivas (NFV), comprimento de estolão (CES), produção de massa seca (MSPA), taxa fotossintética (A), condutância (g) e eficiência do uso da água (EUA). Nas duas avaliações, em ambas as disponibilidades hídricas, verificaram-se correlações positivas e significativas (P<0,01) entre CES e NFV (r≥0,51), assim como entre AF e EST (r≥0,62). O acesso 1 foi o mais tolerante ao estresse hídrico, apresentando maior produção de MS sob alta disponibilidade hídrica, e uma das maiores produções de MS sob déficit hídrico. As características NFV e A poderiam ser usadas para a seleção precoce de plantas de trevo branco para tolerância ao estresse hídrico.


The white clover is a forage legume very important for use in temperate pastures in Rio Grande do Sul. However it is more sensitive to water deficit in the soil than other perennial legumes, presenting a lack of persistence in the summer. With the objective of determining morphophysiological traits related to the answer to the water deficit of this species, an experiment was carried out in greenhouse, which evaluated the effect of water availability (90 and 40 percent of the soil moisture field capacity) on seven genotypes belonging to the white clover core collection from the United States Department of Agriculture (USDA). The experimental design used was completely random, with four repetitions, totaling 56 pots. Two evaluations were made, at 130 and 196 days after the sowing, when several traits were measured. The analysis of the results indicated that the water deficit had an outstanding effect on most of the variables, such as plant height (PH), foliar area (FA), number of alive leaves (NAL), length of primary stolon (LPS), dry matter of the aerial part (DMAP), photosynthesis rates (A), conductance (g), and water use efficiency (WUE). In both evaluations, regardless the water availability, there were positive and highly significant correlations (P<0.01) between LPS and NAL (r≥0.51), as well as between FA and PH (r≥0.62). Therefore, the best access under stress was the 1, which presented larger productions of DMAP at the 90 percent level and one of the most productions of DMAP under stress. The NAL and the A are morphological traits that could be used for early white clover selection for tolerance to water stress.

Nesta revisão são abordados o papel dos gametas não reduzidos (gametas 2n), ou seja, aqueles com o número somático de cromossomos, na origem dos poliplóides, e sua utilização no melhoramento genético de plantas. Aceita-se que a origem dos poliplóides na natureza tenha ocorrido pela fusão de gametas não reduzidos e não pela duplicação somática. Esses gametas são formados por uma não redução cromossômica durante a meiose, que pode ocorrer de duas formas: na meiose I, pela RPD (restituição na primeira divisão), ou na meiose II, pela RSD (restituição na segunda divisão). Normalmente, são encontrados em baixíssimas freqüências em populações naturais, com notáveis exceções, e sua formação é influenciada por fatores genéticos e ambientais. A detecção dos gametas 2n, mais estudados na meiose masculina do que na feminina, é mais freqüentemente feita pela medida de grãos de pólen, ou pela presença de díades e tríades ao final da meiose II. No melhoramento, são utilizados basicamente de duas formas: na poliploidização sexual, uni ou bilateral, que mantém, no poliplóide, a heterozigose que é perdida durante a duplicação somática de cromossomos, e como uma ponte para transferir genes desejáveis de espécies silvestres diplóides para cultivadas poliplóides, como, por exemplo, no caso da alfafa. A manipulação de gametas não reduzidos deve ser considerada como uma ferramenta importante para o melhorista.


The role of unreduced (2n) gametes, those with the somatic chromosome number, in the origin of polyploids and in plant breeding are reviewed. It is accepted that polyploids originated in nature rather by fusion of unreduced gametes than by somatic duplication. These gametes are formed by meiotic non-reduction, in the first division restitution (FDR) or in the second division restitution (SDR). They are normally found in natural populations in very low frequencies, with some remarkable exceptions, and are under genetic and environmental influence. Detection of 2n gametes, which are much more studied in male meiosis, is mainly accomplished by pollen grains size and detection of dyads and tryads at the end of meiosis II. Unreduced gametes are utilized in plant breeding in two ways: in uni or bilateral polyploidization, which maintains in the newly formed polyploid the heterozygosis lost through somatic duplication, and as a bridge to transfer desirable genes from wild diploid species into the cultivated polyploid genepool, by example in alfalfa. The manipulation of unreduced gametes should be considered as an important tool in plant breeding.
